Abstract: The mechanical properties of microenvironments in our body vary over a broad range and these physical cues can be as important for cells as traditional biochemical ones. Especially striking is this mechano-sensitivity in human mesenchymal stem cells (hMSCs) where a systematic variation of the Young’s elastic modulus E of the substrate impacts lineage differentiation.
I will discuss the effects of mechanics on the structure and formation of the cellular cytoskeleton using well-defined extracellular matrix (ECM) models. Using our toolbox Filament-Sensor 2.0 for a quantitative and unbiased analysis of acto-myosin filaments of hMSCs by an order parameter S reveals that stress fiber structure and dynamics are early morphological markers of mechano-guided differentiation. The cytoskeleton structure itself also dictates the shape of the nucleus and lends support to a direct mechanical matrix – stress fibers – nucleus pathway.
